Specimen Preparation

Transport: Deliver the specimen to the laboratory as soon as possible.

Refrigeration: If a delay is necessary, refrigerate the specimen for up to 24 hours. Refrigeration is also recommended if the sample cannot be delivered within 4 hours, to prevent bacterial overgrowth.

Temperature: Specimens for bacterial culture should be transported at room temperature, but refrigeration is acceptable for delays up to 24 hours.

Interferences

Bacterial overgrowth: Leaving the sample at room temperature for more than 4 hours can lead to bacterial overgrowth, which may affect results.

PCR inhibition: Certain substances in the urine can inhibit the PCR reaction. This is why proper collection and prompt delivery are critical to avoid a “”PCR inhibition present”” result.

Contamination: External contamination from skin flora or improperly collected samples can lead to inaccurate results.

Misc Instructions

Patient instructions: Instruct the patient to cleanse the genital area before collection, especially for a clean-catch midstream sample.

Sample type: Midstream clean-catch is preferred for a clean sample. The first voided urine of the morning is also often ideal.

Patient identification: Label the container clearly with the patient’s full name, date of collection, and time of collection.

Communication: Clearly communicate with the patient about the collection procedure to ensure they provide a proper sample.

Clinical information: Ensure the requisition form contains all necessary information, including the patient’s clinical diagnosis, to aid the laboratory in correctly interpreting the results.
